Estriol is a natural estrogen with the chemical name 1,3,5 (10) – triene-3,16 α, 17 β – triol, molecular formula C18H24O3, and molecular weight 288.38 g/mol.
The structure of Estriol is based on an estradiol skeleton, with one hydroxyl group (- OH) on each of the 3, 16, and 17 carbon atoms. This makes Estriol highly water-soluble and easier to metabolize and excrete compared to other estrogens.
Chemical properties of Estriol

This crystal is insoluble in water, slightly soluble in ethanol (1:500), ether, chloroform, dioxane, and vegetable oil, and easily soluble in pyridine and hydroxide alkaline solutions.
biosynthesis
Estriol is mainly synthesized by the ovaries, placenta, and adrenal cortex. In non pregnant women, Estriol is a metabolite of estradiol and estrone, produced through the 16 α – hydroxylation pathway with lower yields.
However, during pregnancy, the placenta becomes the main site for Estriol synthesis: dehydroepiandrosterone (DHEA) produced by the fetal adrenal gland is hydroxylated in the fetal liver and subsequently converted to Estriol in the placenta.
Therefore, the levels of Estriol in the blood and urine of pregnant women are significantly elevated, which can serve as important indicators of fetal placental unit function.
Metabolism and excretion
Estriol is metabolized in the liver through a combination reaction of glucuronidation and sulfation, forming a water-soluble complex that is excreted from urine through the kidneys. Its half-life is relatively short, which is related to weaker estrogenic activity and rapid clearance characteristics.
