What is Nedaplatin?
Nedaplatin (CAS 95734-82-0, molecular formula C₂H₈N₂O₃Pt, molecular weight 303.18) is a second-generation platinum-based antitumor drug developed by Yamanouchi Pharmaceutical Co., Ltd. in Japan and first approved for marketing in Japan in 1995. Its chemical name is cis-diammineglycolatoplatinum(II). It appears as a pale yellow to yellow powder and is soluble in water (13.6 mg/mL) and DMSO, exhibiting better water solubility than first-generation platinum compounds.
As a platinum-based compound, nedaplatin was designed to combine the advantages of cisplatin and carboplatin—retaining cisplatin’s antitumor activity while reducing its nephrotoxicity and gastrointestinal toxicity. Administered intravenously, it is hydrolyzed and activated upon entering cells. Its active metabolites preferentially target the N7 positions of guanine and adenine in DNA, forming intrastrand and interstrand crosslinks that block DNA replication and ultimately induce apoptosis in tumor cells. Nedaplatin also exhibits radiosensitizing effects by making single-strand DNA breaks more likely to form lethal double-strand breaks, thereby enhancing the efficacy of radiotherapy.
Nedaplatin has been used in the treatment of various solid tumors, including head and neck cancers, non-small cell lung cancer, small cell lung cancer, esophageal cancer, ovarian cancer, and cervical cancer. Its dose-limiting toxicity is myelosuppression (particularly thrombocytopenia), although nephrotoxicity and gastrointestinal side effects are significantly lower than those of cisplatin. Moreover, nedaplatin may still be effective in patients resistant to cisplatin, a characteristic that gives it unique value in research on platinum resistance.
