Applications
Polyethylene glycol is a polymer derived from the hydrolysis of ethylene oxide, which is non-toxic and non irritating, and widely used in various pharmaceutical formulations. Low molecular weight polyethylene glycol has relatively high toxicity, and overall, the toxicity of diols is quite low. Local application of polyethylene glycol, especially mucosal administration, can cause irritating pain. In topical cleansers, this product can increase skin flexibility and has a moisturizing effect similar to glycerin. High doses taken orally may cause diarrhea. In injections, the maximum concentration of polyethylene glycol 300 is about 30% (V/V), and hemolysis can occur at concentrations greater than 40% (V/V).
Melting promoter
An excellent chemical promoter for protoplast fusion. It is a polymer of ethylene glycol with the molecular formula HOCH2 (CH2O · CH2) nCH2OH. It is a white, slightly yellowish waxy solid that is soluble in water and has a slight odor. There are a series of polymers with different relative molecular weights, and those with relative molecular weights between 200 and 6000 can be used as melting promoters, but those with a relative molecular weight of 1000 are better (in liquid form at this time). PEG has strong water solubility. In liquid media, the ether bonds on its molecular surface carry weak negative charges. With the participation of Ca2+ions, positively charged surface proteins or negatively charged glycoproteins can be connected through Ca2+bridges, leading to cell aggregation and fusion. In 50% PEG, the disappearance of free water can lead to cell dehydration, resulting in changes in plasma membrane structure and cell fusion.
The advantages of using PEG in cell fusion are:
① Strong universality, suitable for various cells of animals, plants, and microorganisms.
② It is easier to prepare and control than Sendai virus and other viruses.
③ Stable activity and easy to use.
The disadvantages are:
① The effective concentration range is relatively narrow (50% to 55%), but it is more toxic to cells at this time.
② The fusion process of cells cannot be observed under a microscope.
③ The frequency of inducing hybrid cells is relatively low (about 1 × 10-5).
In addition to being used as a melting promoter, it can also be used to promote yeast uptake of exogenous transformed DNA, and can be used as a precipitant in clinical chemistry, as well as a osmoprotectant for serum and other aqueous solutions or suspensions.
FAQ
The number indicates the average molecular weight. PEG 400 is a liquid used in formulations requiring solubility and permeability enhancement. PEG 1000 is a soft wax, often used as a base in solid dosage forms. PEG 4000 and 6000 are solid flakes or powders used for tablet binding, DNA precipitation, and as an excipient in solid formulations. Higher molecular weight grades generally have higher melting points and lower toxicity.
PEG is a versatile material. In pharmaceuticals, it is used as an excipient, tablet lubricant, and protein-modifying agent. In cosmetics, it serves as a humectant and emulsifier. In industry, it is used as a lubricant, dispersant, and softener. In research, it is essential for DNA precipitation, cell fusion, and nucleic acid hybridization
We supply ≥99% purity across multiple molecular weight grades. Packaging options include: 500g, 1kg, 5kg, and 25kg containers. Custom packaging is available upon request. The material is supplied in sealed containers to prevent moisture uptake.
Store in a sealed container in a cool, dry, well-ventilated area. Protect from moisture, as PEG is hygroscopic. Under proper storage conditions (ambient temperature, sealed), the product remains stable for 12–24 months. Keep away from open flames—PEG is combustible, with flash points of 171–260°C depending on molecular weight.
