Tegafur (CAS 17902-23-7): A Comprehensive Introduction

Tegafur, with the CAS number 17902-23-7, is a significant compound in the field of pharmaceuticals and medicine. It is an orally active chemotherapeutic agent that has been widely used in the treatment of various cancers. This article will provide a detailed introduction to Tegafur, including its properties, uses, applications in different fields, and preparation methods.

Properties of Tegafur

Tegafur is a white to off – white crystalline powder. Its chemical name is 1-(tetrahydrofuran-2-yl)-5-fluorouracil. The molecular formula of Tegafur is C₈H₉FN₂O₃, and its molecular weight is approximately 200.17 g/mol. It has a melting point in the range of 162 – 163 °C. Tegafur is sparingly soluble in water, but it can dissolve in organic solvents such as ethanol, acetone, and chloroform. The compound is relatively stable under normal storage conditions, but it should be protected from light and moisture to prevent degradation.

Mechanism of Action

Tegafur acts as a prodrug of 5 – fluorouracil (5 – FU). After oral administration, Tegafur is gradually converted into 5 – FU in the body through a series of enzymatic reactions. 5 – FU is an antimetabolite that interferes with the synthesis of nucleic acids. It inhibits the enzyme thymidylate synthase, which is essential for the synthesis of thymidine, a key component of DNA. By blocking thymidylate synthase, 5 – FU prevents the proper replication and transcription of DNA, leading to the death of rapidly dividing cells, such as cancer cells.

Uses of Tegafur

Anticancer Therapy

Tegafur is primarily used in the treatment of various types of cancers. It has shown efficacy in the treatment of gastrointestinal cancers, including gastric cancer, colorectal cancer, and pancreatic cancer. In gastric cancer, Tegafur – based chemotherapy regimens have been used as adjuvant therapy after surgery to reduce the risk of recurrence. For colorectal cancer, it can be used in combination with other chemotherapeutic agents to improve the survival rate of patients. In pancreatic cancer, although the prognosis is generally poor, Tegafur may be part of palliative chemotherapy to relieve symptoms and slow down the progression of the disease.

It is also used in the treatment of breast cancer. In some cases, Tegafur can be used as a second – line or third – line treatment option when other therapies have failed. Additionally, Tegafur has been investigated for its potential use in the treatment of head and neck cancers, bladder cancer, and ovarian cancer, although its use in these areas may be more limited compared to gastrointestinal and breast cancers.

Combination Therapy

Tegafur is often used in combination with other chemotherapeutic agents or radiotherapy to enhance its anticancer effects. For example, it can be combined with cisplatin, a platinum – based chemotherapy drug. The combination of Tegafur and cisplatin has been shown to have synergistic effects in killing cancer cells. Cisplatin works by cross – linking DNA, which further disrupts DNA replication and repair processes. When used in combination with Tegafur, which inhibits thymidylate synthase, the overall anticancer activity is enhanced.

Another common combination is Tegafur with leucovorin. Leucovorin is a folinic acid derivative that can enhance the binding of 5 – FU (derived from Tegafur) to thymidylate synthase, thereby increasing the inhibitory effect on DNA synthesis. This combination has been widely used in the treatment of colorectal cancer and has shown improved clinical outcomes.

Applications in Different Fields

Medical Research

In medical research, Tegafur is used as a tool compound to study the mechanisms of cancer cell growth and proliferation. Researchers can use Tegafur to investigate the response of different cancer cell lines to 5 – FU – based chemotherapy. By studying the effects of Tegafur on cell cycle progression, apoptosis, and gene expression, scientists can gain a better understanding of the molecular mechanisms underlying cancer development and treatment resistance. Additionally, Tegafur can be used in pre – clinical studies to evaluate the efficacy and safety of new combination therapies or treatment strategies.

Pharmaceutical Industry

In the pharmaceutical industry, Tegafur is used in the formulation of various anticancer drugs. It can be formulated into tablets, capsules, or injection solutions. Pharmaceutical companies need to ensure the quality and stability of Tegafur during the manufacturing process. They also conduct extensive clinical trials to evaluate the efficacy and safety of Tegafur – based drugs in different patient populations. Moreover, the development of new drug delivery systems for Tegafur is an area of active research. For example, liposomal or nanoparticle – based delivery systems can improve the bioavailability of Tegafur and target it specifically to cancer cells, reducing side effects on normal tissues.

Preparation Methods of Tegafur

Classical Synthetic Route

The classical synthesis of Tegafur involves the reaction of 5 – fluorouracil with 2,3 – dihydrofuran in the presence of a suitable catalyst. The reaction typically occurs under reflux conditions in an organic solvent such as dimethylformamide (DMF). The catalyst used is often a base, such as potassium carbonate or sodium hydroxide. The reaction proceeds through a nucleophilic substitution mechanism, where the nitrogen atom of 5 – fluorouracil attacks the electrophilic carbon atom of 2,3 – dihydrofuran, followed by ring – opening and cyclization reactions to form Tegafur.

After the reaction is completed, the crude product is usually purified by recrystallization from a suitable solvent, such as ethanol or a mixture of ethanol and water. The recrystallization process helps to remove impurities and obtain pure Tegafur with a high degree of crystallinity.

Modified Synthetic Approaches

In recent years, several modified synthetic approaches have been developed to improve the yield and purity of Tegafur. One approach is to use microwave – assisted synthesis. Microwave irradiation can significantly accelerate the reaction rate, reducing the reaction time from several hours to a few minutes. This not only improves the efficiency of the synthesis but also reduces the energy consumption. Another modified approach is the use of solid – phase synthesis. In solid – phase synthesis, 5 – fluorouracil is immobilized on a solid support, and the reaction with 2,3 – dihydrofuran is carried out on the solid phase. This method allows for easier separation of the product from the reaction mixture and can be easily automated for large – scale production.
